In Media Server, there's also another setting to pay attention to: D MA Compatability -> Don't not check "Enable Video Transcoding". To get that to work, however, you also need to activate a setting in Media Server: Browsing Settings -> Check the box "Integrate Video Station database into Media Server". I have 3 different types of Roku, and it worked for all of them with MKV files (H264) and MP4 files. Just recently purchased a Synology NAS DS218+ and was able to successfully connect it to the Roku with Roku Media Player. I'm hoping another Synology user using the Synology Media Player can chime in.
It's possible that this is a Synology issue (permissions?). I found an older thread (~ year old) here that was similar: I may not know all the details, but I figure it out and get there.
